The complete TROVO workflow for TIL-based immunotherapy: from tumor tissue dissociation through microwell seeding, longitudinal co-culture imaging, functional clone selection, and live cell recovery. Designed for clinical manufacturing contexts where sample scarcity and reproducibility are critical.

Identify and quantify tumor-killing TILs from clinical samples using TROVO’s Z-score-based potency assessment. By comparing each co-culture microwell against 50 matched tumor-only controls, TROVO generates statistically rigorous killer clone metrics from as few as ~10,000 cells — distinguishing responding from non-responding patient samples without predefined markers.

Rank CAR-T and TCR-T constructs by real-time killing kinetics — not surface markers. TROVO tracks cytotoxicity and persistence live over 12 days across thousands of individual co-cultures, separating true persistence clones from short-term killers before any downstream investment. Retrieved clones maintain full phenotypic expression and functionality post-expansion.

TROVO’s per-clone kinetic data can be fit to Lotka-Volterra predator-prey equations to derive a quantitative killing rate constant (k_eff) for each individual clone — separating intrinsic single-cell killing capacity from cooperative “wolf-pack” killing at higher effector densities. In vitro k_eff profiles fitted from 3,000 microcultures (R²>0.95) correlate directly with in vivo tumor control and survival in NSG mouse models.

TROVO is an “all in one” high throughput imaging system capable of monitoring individual B cell IgG secretion over time with the ability to recover cells of interest then. TROVO can print 20k of hydrogel-based microwells on a 6-well plate, allowing microenvironments to be established. B cells can easily be incubated in our microwells with antigen-coated beads or target cells (suspension/adherent) that can be labeled with a secondary antibody to detect secretion within just 2 hours. These sequestered cells can be tracked over time, captured, and recovered for expansion or sequencing.

Optimize live cell enrichment for single-cell RNA sequencing with Enrich TROVO.
